as an owner what responsibilities (or precautions) can be put in place to prevent a potential legal issues arising from 2 managers developing a relationship? One of these managers would be the store manager the other is the assistant manager.

As long as they are both adults and the relationship appears genuine, I would not be very concerned about legal issues.

On the other hand, I would also talk to them about maintaining professional standards in the work place and that if they do not feel that is going to be possible, that one of them should start looking.

I have had this kind of thing happen a number of times over the years without real problems. A couple of times I have lost an employee when they break up… oh well.

I would keep an eye on things from the perspective of employee morale and avoiding favoritism by the manager, but I would not loose sleep over it.
